The Laboratory of Chemical Biology at the KU Leuven works on the development of chemical probes for covalent labeling of proteins as well as methods to study membrane-embedded enzymes. We utilize an interdisciplinary approach with techniques of organic chemistry, biochemistry, cell biology and proteomics to unravel functions of disease-relevant enzymes.
Project
Proteases cut proteins not only during digestion but also in regulatory processes. Lack of precise control may lead to human disease. Some proteases exert a non-catalytic function beside their canonical proteolytic activity. Hence, inhibition is not sufficient to target these proteases in disease context. In this project, we aim to develop small molecules that degrade extracellular or membrane-bound proteases that are implied in cancer. We will do so by targeting them for endolysosomal uptake and destruction in the lysosomes
In this PhD project at KU Leuven, you will be responsible for evaluating lysosome-targeting compounds in cell models, examining lysosomal degradation of targeted proteases and analyzing the effect on cancer cell phenotypes. This project is supported by a second PhD student in organic synthesis in Leuven and one in organic synthesis and biochemistry in Wroclaw.
